What is MK-677?

MK-677 is a long-acting agonist of the ghrelin receptor and stimulates the production of human growth hormone (GH). This spiroindoline sulfonamide is also known as Ibutamoren, MK-677, L-163,191 and was previously referred to as Oratrope.

Scientific justification

In a study by Jacks et al. (1997), published in Endocrinology, 8 beagles were administered oral doses of MK-677. At 0.25 mg/kg, GH levels increased 5.3x, at 0.5 mg/kg 9x, and at 1 mg/kg even 15.8x. The insulin-like growth factor (IGF-1) remained elevated by 30% for 8 hours. GH remained elevated for up to 6 hours, while the increase in cortisol was relatively small.

Function of ghrelin and ghrelin receptors

MK-677 is a GH secretagogue and an agonist of the ghrelin receptor (GHSR). According to Zhang et al., ghrelin activates this receptor to stimulate food intake, GH release, and reward signaling. Ghrelin also affects stress, glucose metabolism, body weight, cognition, gut motility, food cravings, obesity, and pain perception.

The chemical structure of MK-677 mimics the first four N-terminal residues of ghrelin, including the octanoyl group that is essential for binding to the ligand pocket (source).

Research on neuroprotection

In a study published in the International Journal of Molecular Sciences, mice genetically predisposed to overproduction of amyloid β (Aβ) peptide were administered MK-677 for ten days (0.1 / 1 / 3 mg/kg). Especially at 3 mg/kg, increased food intake was observed, but body weight remained the same between the groups.

MK-677 reduced Aβ deposition, neuronal damage, glial changes, and markers of neuroinflammation. There was also less organ damage and improved neuronal survival observed.
