Andarine S-4
What is S-4?
S-4, also known as Andarine, Androxolutamide, or GTx-007, is a Selective Androgen Receptor Modulator (SARM). In a study by Gao et al., S-4 was characterized as an androgen receptor agonist in orchidectomized rats, with specific effects on skeletal muscles, bones, and the pituitary gland.
Effects on muscle mass
In the study, castrated male rats were administered 3 mg/kg or 10 mg/kg S-4 for 8 weeks. Both dosages were effective in restoring muscle mass and strength in both the soleus muscle and levator ani muscle, to levels comparable to those of intact rats (source).
'The treatment had a minimal effect on the weight of the prostate and seminal vesicles (approximately 16–17%), but showed clear androgenic activity and a suppression of LH and FSH via the pituitary gland.'
Effects on bone mineral density
Gao et al. reported a significant increase in bone mineral density in castrated male rats after 8 weeks of treatment with S-4. An additional study by Kearbey et al. investigated female Sprague-Dawley rats after ovariectomy. It was found that S-4 not only maintained cortical bone volume and bone density but also increased bone strength and reduced body fat (source).
The researchers concluded that S-4 may reduce the risk of fractures by strengthening both muscle and bone structure.
